STIM1 (Stromal Interaction Molecule 1; GOK) is an essential and conserved component of store-operated Ca2+ channel function. It is a Ca2+ sensor that activates CRAC channels and migrates from the Ca2+ store to the plasma membrane (STIM1 translocates from the endoplasmic reticulum to the cell membrane in response to a depletion of intracellular calcium). STIM1 is a typical transmembrane protein with an extracellular N-terminal domain and a cytosolic C-terminal domain. The protein is highly hydrophobic with only a short region of hydrophobicity that likely represents the transmembrane region. The mouse STIM1 homologue (Stim1) was identified independently as a stromal-cell-derived gene product that binds to the surface of haematopoietic cells, particularly pre-B lymphoid cells, and can promote subsequent clonal expansion. Murine Stim1 is 96% identical with human STIM1 in amino acid sequence, and maps to a region of mouse chromosome 7 syntenic with human 11p15.5. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilutions: Western Blot: Reducing and non-reducing conditions. Optimal dilutions to be determined by the researcher. Positive Control: RBL rat basophilic leukemia cell line Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
