SorLA (Sorting protein-related receptor containing LDLR class A repeats; also LR11 and SorL1) is a 250-330kD, monomeric member of the SORL1 family of proteins. It is expressed on the cell surface (10%) and in Golgi/ER membranes (90%) of neurons and vascular smooth muscle cells. SorLA binds apoE-rich lipoproteins plus the 39kD RAP. It also acts as an intracellular sorting receptor for APP. In this context, it blocks transport of APP into compartments that exhibit b-secretase activity, thus interfering with Ab peptide generation. Human SorLA is a type I transmembrane glycoprotein 2214aa in length. It contains a 28aa signal sequence, a 53aa propeptide, and a 2133aa mature molecule. The mature molecule contains a 2056aa extracellular domain (ECD) (aa82-2137) and a short 56aa cytoplasmic tail. The ECD is highly modular, containing five BNR/Asp-box repeats (aa136-573), five LDLR class B repeats (aa 800-1013), one EGF-like domain (aa1026-1072), 11 LDLR class A domains (aa1076-1551) and six FN type III repeats (aa1554-2023). Multiple potential splice variants exist. There are alternative start sites at Met1057 and Met1155, plus a 63aa substitution for (aa1-1153) and a 19aa substitution for (aa1-1404). Over (aa82-367, human SorLA shares 93% aa identity with mouse SorLA. Applications: Suitable for use in Immunofluorescence, Western Blot, ELISA and Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunofluorescence: 1:200-1:1000 ELISA: 1:10,000 Western Blot: 1:500-1:2000 Immunohistochemistry (Formalin fixed paraffin embedded): 1:200 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
