Smac/DIABLO is a mitochondrial protein that is released along with cytochrome c during apoptosis and activates cytochrome c/Apaf-1/caspase-9 pathway. Smac/DIABLO binds to IAPs (Inhibitor of Apoptosis Proteins) and neutralizes the inhibitory effect of IAPs. Analysis of the structural basis of Smac/DIABLO has revealed that the N-terminal aa are required for Smac/DIABLO to bind to IAPs and activation of caspases. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.5-4ug/ml Immunohistochemistry: 5-20ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
