Human CD70 is not present on resting lymphocytes. CD70 is expressed after activation on about 20% of T cells and on about 70% of B cells. CD70 is a ligand for CD27 and it is also a member of the tumor necrosis factor gene family. Applications: Suitable for use in ELISA, Flow Cytometry, Immunocytochemistry and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 1:50 (or use 1.6ul of concentrated stock per 5x10e5 cell test). Optimal dilutions to be determined by the researcher. FACS Protocol: 1. Five x 10e5 cultured Raji human tumor cells were washed and incubated for 45 minutes on ice with 80ul of C2425-01P at a 1:50 dilution (10ug/ml). 2. Cells were washed three times, fixed and analyzed by FACS. 3. Cells stained positive with a mean shift of 1.36 log10 fluorescent units when compared to a I1904-79D IgG1 Murine Negative Control (PE) at a similar concentration. 4. Binding was blocked when pre-incubated for 10 minutes with 20ul of 0.5mg/ml of C2425-01L. Storage and Stability: May be stored at 4 degrees C before opening. DO NOT FREEZE! Stable at 4 degrees C as an undiluted liquid. Dilute only prior to immediate use. Stable for 12 months at 4 degrees C. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. PE conjugates are sensitive to light.
