Human CD62P appears on the surface of activated endothelial cells or platelets and is likely involved with regulating the recruitment of leukocytes to areas of inflammation or infection. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:50. or use 1.6ul undiluted antibody per 5x10e5 cells per test Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: Platelet rich plasma from heparinized blood was activated with 1U/ml Thrombin in the presence of citrate for 10 minutes. Platelets were fixed in 1% buffered formaldehyde, washed twice and pre incubated 5 minutes with 20ul of human IgG at 250ug/ml (to block non specific binding) after which they were incubated 45 minutes on ice with 80ul of C2415-55J at a dilution factor of 1:50 (10ug/ml). Cells were washed three times, fixed and analyzed by FACS. A net 22% of the gated population stained positive with a mean shift of 1.18 log10 fluorescent units when compared to a mouse IgG1/R-PE negative control at a similar concentration. Binding was blocked when pre incubated 10 minutes with 20ul of 0.5 mg/ml C2415-55J. Storage and Stability: May be stored at 4 degrees C before opening. DO NOT FREEZE! Stable at 4 degrees C as an undiluted liquid. Dilute only prior to immediate use. Stable for at least 12 months at 4 degrees C. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. PE conjugates are sensitive to light.
