The human CD1 genetic locus consists of five closely related genes. Three of the gene products (CD1a, CD1b and CD1c) have been officially clustered using antibodies and are expressed on cortical thymocytes, a subset of B cells, and on most professional antigen presenting cells. CD1 molecules have a specialized lipid antigen-presenting function and it is unlikely that they present conventional peptide antigens. CD1 may also be involved in costimulation. Applications: Suitable for use in Flow Cytometry and Immunohistochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:50 (10ug/ml) 80ul labels 5x10e5 human Molt-4 cells Immunohistochemistry: frozen sections Optimal dilutions to be determined by the researcher. Fluorescein/ IgG molar ratio: 9.4 Flow cytometry Protocol: 1. Incubate 5 x10e5 cultured Molt-4 cells on ice for 45 minutes with 80ul of C2252-03F at a 1:50 dilution (10ug/ml). 2. Washed cells 3 times, fixed and analyze by FACS. 3. Cells stained positive with a mean shift of 0.84 log10 fluorescent units when compared to a Mouse IgG1/FITC negative control at a similar concentration. 4. Binding was blocked when cells were pre-incubated with 20ul of 0.5mg/ml of unconjugated CD1c antibody. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. FITC conjugates are sensitive to light.
