Human CD134L is a type II membrane protein with homology to TNF which is expressed on activated B cells and activated endothelial cells. CD134L binds to CD134 present on activated T cells, providing a costimulatory signal (1). This pathway seems to be more effective for costimulation of CD4+ helper T cells than for CD8+ effector T cells (4). Blockade of this interaction in mouse abrogated immunological effects in several models of immflamation and rejection (5,6,7,8). Applications: Suitable for use in Flow Cytometry and ELISA. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:50 labels 5x10e5 HUVEC cells Optimal dilutions to be determined by the researcher. FACS Protocol: 1. Cultured human umbilical cord vein endothelial cells (HUVEC) were washed two times. 2. Five x 10e5 cells per tube were incubated 45 minutes on ice with 80ul of C2515-40D at 10ug/ml. 3. Cells were washed three times, fixed and analyzed by FACS. 4. A net 56% sub population of the cells stained positive with a mean shift 0.95log10 fluorescent units when compared to a Mouse IgG1/R-PE at a similar concentration. 5. Binding was 86% blocked when cells were pre-blocked 10 min with 20ul C2515-40A. Storage and Stability: May be stored at 4 degrees C before opening. DO NOT FREEZE! Stable at 4 degrees C as an undiluted liquid. Dilute only prior to immediate use. Stable for at least 12 months at 4 degrees C. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. PE conjugates are sensitive to light.
