Human CD117 (c-kit) is a transmembrane protein with receptor tyrosine kinase capacity and serves as the receptor for steel factor (SLF). CD117 is expressed on almost all hematopoietic progenitor cells and receptor/ligand interaction is crucial for development of hematopoietic stem cells. Applications: Suitable for use in Flow Cytometry. Does not block binding of SCF. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:50, 80ul/test Optimal dilutions to be determined by the researcher. Protocol: Five x 10e5 cultured HEL human tumor cells were washed and incubated 45 minutes on ice with 80ul of C2448-03K at a 1:50 dilution. Cells were washed three times, fixed and analyzed. Cells stained positive with a mean shift of 1.4 log10 fluorescent units when compared to a mouse IgG1/PE negative control at a similar concentration. Binding was 93% blocked when cells were pre incubated 10 minutes with 20ul of 0.5mg/ml C2448-03K. Storage and Stability: May be stored at 4 degrees C before opening. DO NOT FREEZE! Reconstitute with 1ml sterile dH2O. Stable at 4 degrees C as an undiluted liquid. Dilute only prior to immediate use. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. Protect from light.
