SI-CLP is a secretory protein which is up-regulated in response to interleukin-4 and dexamethasone in activated macrophages. SI-CLP is also expressed by T- and B-lymphocytes, and by cells of monocytic and epithelial origin. SI-CLP is the ligand for stabilin-1, an intracellular sorting receptor which sorts SI-CLP into late endosomes and secretory lysosomes in activated macrophages.
Applications:
Suitable for use in Flow Cytometry and Western Blot. Other applications not tested.
Recommended Dilution:
Flow Cytometry: 1:10-1:100 Use 10ul of the suggested working dilution to label 1x10e6 cells in 100ul. Membrane permeabilization is required for this application.
Western Blot: Detects bands of approximately 33 and 39kD in U937 cell lysates.
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
