The S. cerevisiae silent information regulator 2 (Sir2) protein is an NAD+-dependent histone deacetylase (HDACs) that plays a critical role in transcriptional silencing, genome stability, and longevity (1-2). Part of the Sirtuin family (Sir2, Sir3, Sir3), yeast Sir2 is the founding member of Class III HDACs. A human homologue of Sir2 , SIRT1, regulates the activity of the p53 tumor suppressor and inhibits apoptosis (3-4). Applications: Suitable for use in Flow Cytometry, Western Blot, Immunoprecipitation, Immunohistochemistry and Immunocytochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:30 Western Blot: 1:1000 MW: 110kD Immunohistochemistry: 1:50 Immunocytochemistry: 1:100 Immunoprecipitation: 1:40 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Manufactured incorporating RabMAb(R) technology under Epitomics US patents, No 5,675,063 and 7,429,487, owned by Abcam.
