The retinoblastoma gene product (Rb) is a tumor suppressor protein and acts as cell cycle regulator (1). Recently, Rb has been shown to play a role in apoptosis. During apoptosis, Rb is subjected to cleavage by caspase-3 like protease. Caspase-3 cleaves Rb into 48 and 68kDa fragments (2-4). Cleavage of Rb would make it functionally inactive, causing release of E2F-1. The E2F-1 then may promote apoptosis (5).
Applications:
Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilution:
Optimal dilutions to be determined by the researcher.
Positive Control: camptothecin-treated HL60 (24 hr)
Storage and Stability:
May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 3 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
