RPE65 is a major protein of the retinal pigment epithelium (RPE). RPE65 is essential for the regeneration of rhodopsin in the visual cycle. Mutations in RPE65 are responsible for certain forms of autosomal recessive severe retinal dystrophy, including Leber Congenital Amaurosis (LCA). Loss of function results in a block in retinoid processing required for the synthesis of 11-cis retinal and the accumulation of retinyl esters in the RPE. Applications: Suitable for use in ELISA, Western Blot, Immunohistochemistry, Immunofluorescence and Flow Cytometry. Other applications not tested. Recommended Dilution: Western Blot: 1:5000-1:10,000, recognizes a band at ~65kD representing bovine RPE65 protein. Also been tested on recombinant human protein in COS7 cells. Immunohistochemistry (Frozen): 1:250 on murine retinal sections Immunohistochemistry (paraffin): 1:250 Immunofluorescence (IC): 1:10-1:2000 Optimal dilution determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
