Cell cycle progression is regulated by cyclins and their cognate Cdks. p21 (WAF1/CIP1) inhibits the activity of each member of the cyclin/Cdk family, and over expression of this protein inhibits the proliferation of mammalian cells. The expression of p21 is inducible by a wide range of stress stimuli. p21 is tightly regulated at the transcriptional level by p53, and probably serves as the effector of p53 cell cycle control. Cellular Localization: Nuclear Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry (paraffin-embedded sections). Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot Store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
