Nuclear factor B (NF-B) encompasses an important family of inducible transcriptional activators that regulate a wide variety of cellular and viral genes (1). The family members include p50, p52, p65 (RelA), c-Rel, and RelB (2). Association with inhibitory proteins of the IkB family retains NF-kB in the cytoplasm. Degradation of IB proteins exposes the nuclear localization sequence (NLS), leading to nuclear translocation and subsequent binding of NF-B to DNA (3). The p105 precursor protein acts as an NFkB inhibitor, retaining Rel subunits in the cytoplasm of unstimulated cells. Upon stimulation by TNFa and IL -1a, degradation of p105 occurs; the Rel subunit is released and translocats to the nucleus (4). Additionally, p105 will be proteolytically processed by the 26S proteasome to produce a smaller, transcriptionally active form (p50) (5). Applications: Suitable for use in Flow Cytometry, Western Blot, Immunohistochemistry, Immunocytochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:30 Western Blot: 1:5,000 Immunohistochemistry: 1:250-500 Immunocytochemistry: 1:250-500 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
