In the Human Midkine ELISA, Calibrators, Quality Controls, and samples are incubated with a rabbit polyclonal anti-human midkine antibody coated in microtiter wells. After two hours of incubation and washing, biotin-labeled polyclonal anti-human midkine antibody is added and incubated with captured midkine for one hour. After a thorough wash, streptavidin-horseradish peroxidase conjugate is added. After one hour of incubation and the last washing step, the remaining conjugate is allowed to react with the substrate H2O2-tetramethylbenzidine. The reaction is stopped by the addition of acidic solution and the absorbance of the resulting yellow product is measured at 450nm. The absorbance is proportional to the concentration of midkine. A standard curve is constructed by plotting the absorbance values against the concentrations of midkine calibrators. The concentrations of unknown samples are determined using this standard curve. Samples: serum, citrate plasma and heparin plasma.
