This assay is a simple two-step procedure performed in a microtiter plate. In the first step samples are incubated with the HDAC assay substrate, allowing deacetylation of the fluorometric substrate. Activator Solution releases a fluorophore from the deacetylated substrate or standard. Quantity: Sufficient reagents for 96 assays per kit. Kit Components Half volume white plate One half volume 96 well white plate. Half volume black plate One half volume 96 well black plate. HDAC Assay Buffer One vial containing 10ml of 25mM Tris pH 8.0, 137mM NaCl, 2.7mM KCl, 1mM MgCl2. Activator Solution One vial containing 200ul of activator solution, 20X stock. Aliquot upon receipt as needed to avoid future freeze-thaw cycles. HDAC Assay Substrate One vial containing 100ul of 5mM substrate in DMSO. HDAC Assay Standard One vial containing 50ul of 1mM unacetylated standard in DMSO. Trichostatin A One vial containing 100ul of 200uM Trichostatin A in DMSO. HeLa Nuclear Extract One vial containing 50ug in 25ul of nuclear extract prepared from human HeLa cells in RIPA buffer. Aliquot upon receipt as needed to avoid future freeze-thaw cycles. Storage: Upon receipt, see individual components for storage conditions. The half volume 96 well plates should be stored at room temperature. All other components are stored at -20 degrees C. Stability: Components are stable for 6 months from date of shipment if stored and handled correctly.
