Type I collagen is the major structural component of extracellular matrices found in connective tissue and internal organs, but is most prevalent in the dermis, tendons, and bone. It is a 300kD molecule composed of two alpha 1(I) chains and one alpha2(I) chain that spontaneously forms a triple helix scaffold when at a neutral pH and 37 degrees C. This phenomenon can be exploited to promote cell attachment, growth, differentiation, migration, and tissue morphogenesis during development. Quality Control: Tested for uniform gelation and for attachment of and spreading of HT-1080 human fibrosarcoma cells. Negative for the presence of mycoplasma, bacteria and fungi. Technical Notes: 1. Typically if the rat tail collagen's concentration goes below 0.4mg/ml, there will be little to no gelation. 2. Higher pH can also interfere with the gelling of this product. Storage and Stability: May be stored at 4 degrees C. For long-term storage, aliquot and store at 4 degrees C. Do not freeze. Aliquots are stable for 6 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
