Human CD94 expression is restricted to natural killer (NK) cells and a small subset of T lymphocytes. CD94 is a type II membrane glycoprotein of the c-type lectin superfamily and essentially lacks a cytoplasmic tail. Applications: Suitable for use in Flow Cytometry and Functional Application. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:100 Functional Application: Antibody inhibits IL-2 dependent proliferation of NK cells. Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: Ficoll prepared human peripheral white blood cells were stimulated by incubating 1 day at 5x10e6 cells/ml in RPMI, 10% FBS media including 5ug/ml Phytohemagglutinin-P. Five x 10e5 cells/tube were then washed and incubated 45 minutes on ice with 80ul of C2442-02B at a 1:100 dilution. Cells were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.09 log10 fluorescent units when compared to a Mouse IgG1 (FITC) Mouse negative control (I1904-75G) at a similar concentration. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. FITC conjugates are sensitive to light.
