Human CD63 is expressed on activated platelets and also on monocytes and macrophages. CD63 contains four hydrophobic transmembrane regions and is a member of the tetraspan family. CD63 appears to be involved with cell adhesion and associates with VLA-3 and VLA-6 integrins.
Applications:
Suitable for use in Flow Cytometry. Other applications not tested.
Recommended Dilution:
Flow Cytometry: 1:50 (or use 1.6ul concentrated stock per 5x10e5 cells)
Optimal dilutions to be determined by the researcher.
Performance:
5x10e5 cultured HPB-MLT cells were washed and pre incubated 5 minutes with 20ul of human IgG at 250ug/ml (to block non specific binding) after which they were incubated 45 minutes on ice with 80ul of C2418-02E at a 1:50 dilution factor (10ug/ml). They were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.12 log10 fluorescent units when compared to a mouse IgG1/FITC negative control at a similar concentration. Binding was blocked when cells were pre-incubated 10 minutes with 20ul of 0.5mg/ml anti-CD63 antibody (Cat. #C2418-02C).
Storage and Stability:
May be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
