Human CD60 antigen is a glycolipid determinant found on about 30% of peripheral blood T cells. The glycolipid determinant is linked to a glycoprotein which can transduce activation signals. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:50; 80ul labels 5x10e5 ficoll prepared human PBL cells. Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. 5x10e5 ficoll prepared human PBL were incubated 45 minutes on ice with 80ul of C2413-79B at a 1:50 dilution (10ug/ml). 2. Cells were then washed three times, fixed and analyzed by FACS. A 39% sub population of the cells stained positive with a mean shift of 2.3 log10 fluorescent units when compared to a Mouse IgM/FITC negative control at a similar concentration. 3. Binding was blocked when cells were pre incubated for 10 minutes with 20ul of 0.5mg/ml anti-CD60b antibody (C2413-79). Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
