Human CD49b is an integrin alpha 2 subunit that forms a heterodimer with beta 1 integrin and functions as an adhesion molecule. Applications: Suitable for use in Flow Cytometry and Immunohistochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:50. Use 80ul per test. Optimal dilutions to be determined by the researcher. FACS Protocol: 1. Five x 10e5 cultured UM-SCC (squamous cell carcinoma) cells were harvested by trypsinization, washed and preincubated 5 minutes with 20ul of 250ug/ml human IgG (To block nonspecific binding). After which, they were incubated 45 minutes on ice with 80ul of C2404-07K at a 1:50 dilution. 2. Cells were washed three times, fixed and analyzed by FACS. 3. Cells stained positive with a mean shift of 1.08 log10 fluorescent units when compared to a Mouse IgG2a/FITC negative control at a similar concentration. 4. Binding was 91% blocked when pre-incubated 10 minutes with 20ul of 0.5ug/ml C2404-07H. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. FITC conjugates are sensitive to light.
