Human CD46 is a cell surface glycoprotein expressed on almost all human cells except erythrocytes. CD46 is involved with regulation of complement activation and is also a measles virus receptor. Applications: Suitable for use in Flow Cytometry and Western Blot. Other applications not tested. Recommended Dilutions: Flow Cytometry: 1:50 (10ug/ml) labels 5x10e5 cells Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Five x 10e5 cultured human HPB-MLT cells per tube were washed and incubated 45 minutes on ice with 80ul of C2401-04A at a 1:50 dilution (10ug/ml) 2. Cells were washed three times, fixed and analyzed by FACS. 3. Cells stained positive with a mean shift of 1.1 log10 fluorescent units when compared to a Mouse IgG2a/FITC negative control at a similar concentration. 4. Binding was blocked when pre incubated 10 minutes with 20ul of 0.5 mg/ml C2401-04. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. FITC conjugates are sensitive to light.
