CD40, a member of the TNFR-SF, is a 45-48kD type I integral membrane glycoprotein whose ligand is CD154. It is expressed on all mature B lymphocytes, but is absent from plasma cells. It is also present on some epithelial and endothelial cells. CD154 binding to CD40 on B cells is important for T cell/B cell interactions and is required for secondary immune responses. Evidence for a role of CD40-CD154 interaction in negative selection and peripheral tolerance has been suggested. CD40 monoclonal antibodies have been reported to stimulate B cell proliferation when costimulated with anti-mu, CD20 monoclonal antibodies or phorbol esters.1-3 Applications: Suitable for use in Flow Cytometry and B cell assays. Other applications not tested. Recommended Dilution: Flow Cytometry: 20ul/10e6 cells Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C before opening. DO NOT FREEZE! Stable at 4 degrees C as an undiluted liquid. Dilute only prior to immediate use. Stable for at least 12 months at 4 degrees C. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. PE conjugates are sensitive to light.
