Human CD27 is a lymphocyte specific member of the tumor necrosis factor (TNF) receptor family and is found primarily on peripheral blood T cells and on a subpopulation of B cells and NK cells. The ligand for CD27 is CD70, which is a member of the TNF ligand superfamily. Applications: Suitable for use in ELISA and Flow Cytometry.Other applications not tested. Recommended Dilution: Flow Cytometry: 1:50 Use 80ul to label 5x10e5 cells. Optimal dilutions to be determined by the researcher. Flow CytometryProtocol: 1. Wash 5x10e5 ficoll prepared human peripheral blood lymphocytes per tube. 2. Preincubated 5 minutes with 20ul of 250ug/ml human IgG (To block non specific binding) 3. Wash and incubated 45 minutes on ice with 80ul of C2379-09R at a 1:50 dilution. 4. Washe three times, fixed and analyzed on a BD FACS Calibur. A net 54% sub population of the cells stained positive with a mean shift of 1.19 log 10 fluorescent units when compared to a Mouse IgG1/FITC negative control at a similar concentration. Binding was 99% blocked when cells were pre incubated 10 minutes with 20ul of 0.5mg/ml C2379-09P. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. FITC conjugates are sensitive to light.
