Human CD23 is a low affinity receptor for IgE (Fc epsilon RII) found primarily on mature B cells and also on eosinophils, monocytes, dendritic cells, platelets, and a subset of thymic epithelial cells. CD23 interacts with CD21. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:50 Immunohistochemistry: Formalin fixed paraffin embedded sections Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Five x 10e5 ficoll prepared human peripheral white blood cells were washed and preincubated for 5 minutes with 20ul of 250ug/ml human IgG (to block non-specific binding) after which they were washed and incubated 45 min on ice with 80ul of C2273-91L at a 1:50 dilution. 2. Wash cells three times, fixed and analyzed using BD FACS calibur. 3. A net 13.6% sub-population of the cells were stained positive with a mean shift of 1.67 log10 flourescent units when compared to a IgG1 Murine (PE) negative control at a similar concentration. 4. Binding was 95% blocked when pre-incubatde 10 minutes with 20ul of 0.5mg/ml C2273-91H. Storage and Stability: May be stored at 4 degrees C before opening. DO NOT FREEZE! Stable at 4 degrees C as an undiluted liquid. Dilute only pror to immediate use. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity.
