Human CD16 is a low affinity receptor for complexed IgG and occurs in two distinct forms, as a glycosyl phosphtidyl inositol (GPI) anchored protein in granulocytes, and as an integral membrane protein on NK cells and macrophages. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Immunohistochemistry: frozen sections. Flow Cytometry: 1:50 for 5x10e5 cells. Optimal dilutions to be determined by researcher. Flow Cytometry Protocol 1. Wash 5 x10e5 cells of ficoll prepared human peripheral blood lymphocytes. 2. Incubate on ice for 45 minutes with 80ul of C2267-33D at 1:50 dilution (10ug/ml). 3. Wash 3 times, fix and analyze by FACS. 4. An 25% sub-population of the cells stained positive with a mean shift of 2.1 log10 fluorescent units when compared to a mouse IgG1(PE) negative control at a similar concentration. 5. Binding was blocked when cells were pre-incubated for 10 min. with 20ul of 0.5 mg/ml anti-CD16 antibody (C2267-33A). Storage and Stability: May be stored at 4 degrees C before opening. DO NOT FREEZE! Stable at 4 degrees C as an undiluted liquid. Dilute only prior to immediate use. Stable for at least 12 months at 4 degrees C. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. PE conjugates are sensitive to light.
