CD154, also known as CD40 ligand and a member of the tumor necrosis factor superfamily of cell surface receptors, is a 33 kD type II transmembrane protein that is expressed as a trimer on the cell surface. It is absent from resting lymphocytes but its expression is rapidly upregulated upon activation. It is present mostly on CD4+ cells but also on a small subpopulation of CD8+ cells and gamma/delta+ T cells. It is also found on activated basophils and mast cells. CD154 binding to CD40 on B lymphocytes is required for secondary immune responses and germinal center formation. This interaction may also play a role in negative selection and peripheral tolerance.1-4 Applications:
