Human CD134 (OX40) is an activation-associated antigen which is predominantly expressed on activated CD4 positive cells. CD134 antigen is a member of the tumor necrosis factor (TNF) receptor family of molecules and may be involved with regulating T cell-dependent B cell proliferation and differentiation. Applications: Suitable for use in Flow Cytometry and ELISA. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:50 (80ul/test) Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. 5x10e5 cultured HPB-MLT cells were washed and incubated 45 minutes on ice with 80ul of C2515-19B at 1:50 dilution factor. 2. Cells were washed three times and fixed and analyzed on a BD FACstar plus. 3. Cells stained positive with a mean shift of 0.9 log10 fluorescent units when compared to a Mouse IgG1/FITC Negative Control at a similar concentration. 4. Binding was 77% blocked when cells were preincubated 10 minutes with 20ul of 0.5mg/ml C2515-19, CD134 (OX40). Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. FITC conjugates are sensitive to light.
