Human CD134L (OX-40 Ligand) is a type II membrane protein with homology to TNF which is expressed on activated B cells and activated endothelial cells. CD134L binds to CD134 present on activated T cells, providing a costimulatory signal (1). This pathway seems to be more effective for costimulation of CD4+ helper T cells than for CD8+ effector T cells (4). Blockade of this interaction in mouse abrogated immunological effects in several models of immflamation and rejection (5,6,7,8). Applications: Suitable for use in Flow Cytometry, ELISA and blocking studies. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:50 (or 1.6ul neat/5x10e5 cells/test) Blocking: Blocks binding of Recombinant CD134 to CD134L on HUVEC. Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Cultured human umbilical cord vein endothelial cells (HUVEC) were harvested by trypsinization. 5x10e5 cells per tube were washed and incubated 45 minutes on ice with 80ul of C2515-40C at a 1:50 dilution (10ug/ml). 2. Washed three times, fixed and analyzed on a BD FACS Calibur A. Binding was 87% blocked when cells were pre-incubated 10 minutes with 20ul of 0.5mg/ml C2515-40A. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
