Human CD13 is a zinc-binding aminopeptidase-N enzyme expressed on the surface of myeloid cells. IL-4 will upregulate expression of CD13 which may play an anti-inflammatory role. Applications: Suitable for use in Flow Cytometry and Immunohistochemistry (paraffin). Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. FACS Protocol: 1. 5 x10e5 cultured THP-1 cells were washed and pre incubated ~ 5 minutes with 20ul of 250ug/ml of human IgG. 2. Incubate 5 x10e5 cells on ice for 45 minutes with 80ul of C2264-01D at 1:50 dilution. 3. Washed 3 times, fixed and analyzed on a BD FACS Calibur. 4.. Cells stained positive with a mean shift of 2.47 log10 fluorescent units when compared to a Mouse IgG2a (PE) negative control at a similar concentration. Binding was 86% blocked when reagent was pre-incubated for 10 minutes with 20ul of 0.5mg/ml C2264-01A. Storage and Stability: May be stored at 4 degrees C before opening. DO NOT FREEZE! Stable at 4 degrees C as an undiluted liquid. Dilute only pror to immediate use. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity.
