BAFF, Recombinant, Human (B cell Activating Factor belonging to TNF Family, BLyS, TALL-1, THANK)

The TNF-related ligand, B cell-activating factor belonging to the TNF family (BAFF), is necessary for normal B cell development and survival, and specifically binds the receptors transmembrane activator and calcium-modulator and cyclophilin ligand interactor (TACI), B cell maturation Ag (BCMA), and BAFF-R.Using specific mAbs, BAFF-R was found to be the predominant BAFF receptor expressed on peripheral B cells, in both humans and mice, and antagonist mAbs to BAFF-R blocked BAFF-mediated costimulation of anti- micro responses. The other BAFF receptors showed a much more restricted expression pattern, suggestive of specialized roles. Recombinant Human B Lymphocyte Stimulator is a single, non-glycosylated polypeptide chain containing 153 amino acids and having a molecular mass of 17,007 Daltons. Source: E. coli Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Met-Ala-Val-Gln-Gly. Biological Activity: Recombinant B Lymphocyte Stimulator is fully biologically active when compared to standard. The activity is determined by the dose-dependent stimulation of IL-8 production by human PBMC. The ED50 for this effect is less than 10ng/ml. Please note: results may vary with PBMC donors. Protein Content: Protein quantitation was carried out by two independent methods: 1. UV spectroscopy at 280. 2. Analysis by RP-HPLC, using a standard solution of BAFF as a Reference Standard. Reconstitution: Reconstitute the lyophilized BAFF in sterile 18MOmega-cm H2O not less than 100ug/ml, which can then be further diluted to other aqueous solutions. Storage and Stability: Lyophilized powder may be stored at 4 degrees C for short-term only. Reconstitute to nominal volume by adding sterile dH2O and store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
